Immunoblotting of Secreted Cytolytic Substance in Culture Media of Acanthamoeba culbertsoni

Abstract

|
|
The cytolytic substance of the pathogenic free-living amoeba is a etiological agent of primary amoebic meningoencephalilis. The molecular weight of the purified cytolytic substance from Acanthamoeba culbertsoni was 33 kD and it was used as antigen to detect the substance from culture media of the amoeba. The enzyme-linked immunoblotting result between SDS polyacrylamide gel electrophoresis of the culture media and the rabbit antiserum showed a major band and 3 minor bands. The major band revealed the presence of a antigen-antibody complex having a molecular weight of 33 kD, indicating that the cytolytic substance of the pathogenic amoeba may be released to the culture media of the amoeba.
